چکیده
Beers, Roland F., Jr. (Johns Hopkins University, Baltimore, Md). Acridine orange binding by Micrococcus lysodeikticus. J. Bacteriol. 88:1249-1256. 1964.-Micrococcus lysodeikticus cells bind acridine orange (AO) reversibly. The adsorption isotherm is consistent with a highly cooperative-type binding similar to that observed with polyadenylic acid. The cells exhibit a strong buffering action on the concentration of free AO which remains constant (1 mug/ml) over a range from 5 to 95% saturation of the cells by AO. The cells stain either fluorescent orange or green. The fraction stained orange is directly proportional to the quantity of dye adsorbed, indicating that these cells bind a fixed amount of AO (10% of dry weight). The green-stained cells contain less than 1% of the AO bound to orange-stained cells. The results suggest that the abrupt increase in amount of AO bound by the orange-stained cells occurs when the concentration of free AO reaches a threshold concentration. Similar results were obtained with Bacillus cereus. Mg increases the free AO concentration and the extent of binding capacity of the cells.

Post-protein binding metal-mediated coupling of an acridine orange-based fluorophore.
The HEW lysozyme (Lys) and the fac-[Re(CO)(3)(H(2)O)(3)](+) complex (1) are used as a simple model system for the description of a new approach to the labelling polypeptides with fluorescent tags. The strategy takes advantage of the reaction of an acridine orange-based fluorophore (AO) with the non-native metal fragment 1 hybridized on the enzyme. A synthetic methodology for the quantitative me...
متن کاملInhibition and activation of polynucleotide phosphorylase by acridine orange.
Preliminary studies by Beers et a2. (1) have shown that the zinc salt of acridine orange can either inhibit or accelerate the rate of polymerization of polynucleotides by polynucleotide phosphorylase of Micrococcus lysodeileticus, depending upon the experimental conditions. We have now confirmed these findings with the zinc-free dye. Some evidence for a reversal of inhibition with degraded ribo...

متن کاملBinding of bacitracin to cells and protoplasts of Micrococcus lysodeikticus.
Bacitracin was tritiated by the Wilzbach technique and purified by carboxymethylcellulose chromatography. Binding studies between [3H]bacitracin and Micrococcus lysodeikticus cells and protoplasts indicated that the cells or protoplasts were saturated at 2 X lo5 molecules per cell. This value correlates well with the number of CS5-isoprenyl pyrophosphate molecules found in the membrane and adds...
متن کاملMolecular cloning and characterization of a new G-type lysozyme gene (Ec-lysG) in orange-spotted grouper, Epinephelus coioides.
Lysozyme acts as an innate immunity molecule against pathogen infection. In this study, a new G-type lysozyme gene with a typical G-type lysozyme domain (designated as Ec-lysG) was cloned and characterized from the orange-spotted grouper, Epinephelus coioides. The full-length Ec-lysG cDNA contains 1419 bp and encodes a 256-residue protein containing a 25-residue signal peptide at the N-terminus...
